¿Quiere usted? or ¿Quieres tú? - Do you want?
¿Quieres tú? = "¿Quieres?" (familiar)
¿Quiere ud.? = ¿Quiere? (formal) (ud. is short for usted)
You don't need the pronouns "tú" or "ud." You can just easily say "¿Quiere?" or ¿Quieres"
Question: When do you use ¿Quiere? or ¿Quieres?? ¿Quiere? is the formal way of asking, "Do you want?". You use ¿Quiere? when speaking to grandparents, the boss, teachers, people you don't know. etc. ¿Quieres? is the familiar way of asking, "Do you want?". You use ¿Quieres? when speaking with children or to friends. When I first started learning Spanish, I used both ¿Quiere? and ¿Quieres? and it was fine.
"tu quieres" or "usted quiere" is the way you tell somebody what they want.
For example:
"Tú quieres la camisa. - You want the shirt." (familiar)"Quieres ir a la playa. - You want to go to the beach." (familiar)
"Usted quiere los pantalones. - You want the pants." (formal) "Quiere comprar un gato. - You want to buy a cat." (formal) I don't know why you would ever want to tell someone what they want, but I was giving you the example just to let you know.
Definition: Intonation is the way your voice changes when you ask a question or a statement.
Also, if you change the Intonation of your voice to a question when saying "Tu quieres la camisa" then are you actually asking the question, "Do you want a shirt?" and not telling someone "You want a shirt." If you are saying "Quieres la camisa"(without the pronoun) and you don't say it like a question, then it could be interpreted as a statement and you are telling the person that they want a shirt.
